I. Background
GLP-1R 与其他四种胰高血糖素受体（GCGR、GLP-2R、GIPR 和 GHRHR）一起属于促胰液素 (B1 类) GPCR 家族，其内源性配体是肽激素。 B1 类 GPCRs 在 N 端具有一个较大的、结构保守的胞外结构域（ECD），由 120-160 个残基组成，形成三层 α-β-β-α 折叠，并由三个层间二硫键稳定。 GLP-1R 的内源性配体是 GLP-1 （7-36）和 GLP-1 （7-37），它们是胰高血糖素原翻译后加工的产物。GLP-1R 主要通过 Gαs/cAMP 通路发出信号，直接导致葡萄糖诱导的胰岛素颗粒分泌；但有证据表明 GLP-1R 可与 Gαq 和其他 G 蛋白偶联；GLP -1R 激活后，C 末端发生磷酸化，进一步募集β-抑制蛋白，导致受体内化和脱敏
